چکیده

The Diaphanous-related formins are effectors for Rho GTPases. Two recent reports have unveiled previously unappreciated activities of Drf3 (mDia2), eliciting cytoskeletal rearrangements at the behest of Cdc42, and of DRF2 (hDia2C), regulating endosome dynamics for RhoD.

Formins as effector proteins of Rho GTPases

Formin proteins were recognized as effectors of Rho GTPases some 15 years ago. They contribute to different cellular actin cytoskeleton structures by their ability to polymerize straight actin filaments at the barbed end. While not all formins necessarily interact with Rho GTPases, a subgroup of mammalian formins, termed Diaphanous-related formins or DRFs, were shown to be activated by small GT...

The small GTPase Rif is an alternative trigger for the formation of actin stress fibers in epithelial cells.

Actin stress fibers are fundamental components of the actin cytoskeleton that produce contractile force in non-muscle cells. The formation of stress fibers is controlled by the small GTPase RhoA and two highly related proteins, RhoB and RhoC. Together, this subgroup of actin-regulatory proteins represents the canonical pathway of stress-fiber formation. Here, we show that the Rif GTPase is an a...

Structural basis of Rho GTPase-mediated activation of the formin mDia1.

Diaphanous-related formins (DRFs) regulate dynamics of unbranched actin filaments during cell contraction and cytokinesis. DRFs are autoinhibited through intramolecular binding of a Diaphanous autoinhibitory domain (DAD) to a conserved N-terminal regulatory element. Autoinhibition is relieved through binding of the GTPase RhoA to the N-terminal element. We report the crystal structure of the di...

A network of conserved formins, regulated by the guanine exchange factor EXC-5 and the GTPase CDC-42, modulates tubulogenesis in vivo.

The C. elegans excretory cell (EC) is a powerful model for tubulogenesis, a conserved process that requires precise cytoskeletal regulation. EXC-6, an ortholog of the disease-associated formin INF2, coordinates cell outgrowth and lumen formation during EC tubulogenesis by regulating F-actin at the tip of the growing canal and the dynamics of basolateral microtubules. EXC-6 functions in parallel...

Diaphanous-related Formin homology proteins

What are Diaphanous-related Formin homology (DRF) proteins? DRF proteins constitute a subfamily of Rho GTPase-binding Formin homology (FH) proteins, first defined after the discovery of conserved FH1 and FH2 domains shared between Diaphanous and Bni1p, required for cytokinesis in flies and yeast, respectively, and Formins, limb deformity genes in mice. As effectors for activated, GTP-bound Rho,...
